V8 Protease, also called Endoprotease Glu-C, is used to selectively cleave proteins for amino acid sequence determination or for peptide mapping. V8 protease cleaves the peptide bonds on the carboxyl side of aspartic and glutamic acid residues when buffered in phosphate. When buffered in ammonium bicarbonate or ammonium acetate, V8 protease only cleaves on the carboxyl side of glutamic acid residues.
